I swear I have been able to set CEPro to be able to start playing a wave file at any given point in the file by just left-clicking the mouse. As it stands, I can select the point, but must hit the play button for it to play at the cue point. In other words..I just want to be able to left click the mouse on the wave at any point and it will start playing from there.

As far as I know it's not possible to do this...just clicking somewhere in the waveform and it starts to play from that point...
But you can make cue points first. Then in the cue list you can choose auto-play. Then a cue will automatically start to play when you select it in the cue list.

The behaviour you describe isn't a Cool Edit feature (maybe you were thinking of another package) but there is a fair bit of customisation which you can do via the keyboard shortcuts (normally alt/k) and by right clicking on each of the transport controls.
